The 24-year-old replaced Pietro Comuzzo as La Viola pushed for another goal moments after Luca Ranieri had pulled one back for the hosts.
Meanwhile, in a dominant first half display by the champions, former Manchester City captain Kevin De Bruyne opened the scoring for Napoli from the spot in the 6th minute.
Eight minutes later, on-loan Manchester United forward Rasmus Hojjlund registered his first goal for the club before Sam Baukema added the third six minutes after the break.
Despite Fiorentina's late attack, Stefano Pioli's men could not match the strength of Antonio Conte's team.
Meanwhile, Lamptey will be hoping to gain more game time after an impressive cameo.
